Sequencing

Precipitation after the sequencing reaction

There are several possible precipitation protocols for sequencing reactions (even one recommended by ABI); this is the one we use now.

  1. Combine 980µl 70%etoh with 20µl 25mM MgCl. This step should be done just before each batch or reactions is processed.
  2. Add 74µl to each sequencing reaction and mix well (vortexing is ok).
  3. Let sit in the dark at room temperature for 20 min.
  4. Spin at 5300rpm for 30 min.
  5. Pour off or draw off liquid (tap our reminder if pouring); allow to airdry or use speedvac (3-4 min.)
